Tetracycline Hydrochloride is a broad-spectrum antibiotic active against Gram-negative and Gram-positive bacteria. It inhibits protein synthesis by binding the bacterial ribosome and blocking aminoacyl-tRNA binding at the A-site, which halts translation. Owing to its reliability and consistent performance, it is widely used for bacterial selection (Tetᴿ) and as a ligand in tetracycline-controlled gene expression systems (Tet-On/Tet-Off) in eukaryotic cells.

Applications

  • Selection of Tetᴿ bacteria (plasmids with tetA/tetR markers). Typical final concentration: 10–20 µg/mL in LB/agar; titrate by strain.

    • Bacterial selection: typically 10–20 µg/mL (LB broth/agar); titrate by strain/plasmid.
    • Tet-On/Tet-Off: start 0.1–1 µg/mL tetracycline/doxycycline and optimize per cell line.
