Q1: Why is RNase A included in the kit?
To remove RNA, ensuring PI binds selectively to DNA.

Q2: What cell number can be used for this assay?
The standard protocol recommends 2–5 × 10⁵ cells per sample. However, higher cell numbers (e.g. up to 1 × 10⁶ cells) may also be used, provided that proper resuspension and homogeneous staining are ensured.

Q3: How should cells be detached prior to staining?
Cells can be harvested using different approaches depending on the cell type, including enzymatic detachment (e.g. trypsinization) or mechanical methods such as a cell scraper.

Q4: Are variations in centrifugation conditions acceptable?
While the standard protocol specifies centrifugation at 500 × g, slight variations in centrifugation speed may be used as long as cell integrity is maintained and pellets are not disrupted during processing.
