FastPANGEA™ High Fidelity DNA Polymerase is a second-generation, proofreading enzyme that delivers extreme accuracy, speed, and robustness for all PCR applications.
With an error rate at least 50× lower than standard Taq DNA Polymerase, it amplifies long and complex templates (up to 15 kb) with blunt-end product generation — ideal for high-precision molecular cloning and sequencing workflows.

As a second-generation high-fidelity enzyme, it provides a unique balance of speed, robustness, and ultra-low error rate, making it suitable for both routine and demanding PCR applications.

This advanced enzyme possesses 5′→3′ DNA polymerase and 3′→5′ exonuclease (proofreading) activities, ensuring accurate nucleotide incorporation and minimal mutation rate.
It efficiently amplifies long DNA fragments (10–15 kb) and GC-rich or structurally difficult templates with extension times of only 15–30 seconds per kb.

The resulting PCR products are blunt-ended, enabling direct cloning into blunt-end or GC-cloning vectors.

Unit definition:
One unit is defined as the amount of enzyme that incorporates 10 nmoles of dNTP’s into acid-insoluble form in 30 minutes at 75 °C under assay conditions: 25 mM TAPS-HCl, pH 9.0 (at 25 °C), 100 mM KCl, 1.5 mM MgCl2, 1 mM Beta-mercaptoethanol, 200 μM each dNTP and 10 μg activated calf thymus DNA in 50 μl.

      We incorporated this high-fidelity polymerase into our cloning workflow for amplifying several constructs above 3 kb. The enzyme is supplied with the corresponding buffer, which simplifies reaction setup and reduces variability between users. Cycling conditions were easy to adapt from our previous protocol, and amplification profiles were clean on agarose gels. It fits well in projects where sequence accuracy is critical before Sanger verification.
